1. INTRODUCTION
Thyroglobulin (TG) is a 660 kDa, dimeric glycoprotein produced by the thyroid gland and involved in the storage and synthesis of thyroid hormones. It is the precursor of the thyroid hormones 3,3’,5-triiodo-L-thyronine (T3) and L-thyroxine(T4).The enzyme thyroperoxidase (TPO) promotes iodination of tyrosine residues in thyroglobulin molecules, formingmonoiodotyrosine (MIT) and diiodotyrosine (DIT). TPO further catalyzes the intramolecular coupling of two molecules ofdiiodotyrosine to produce tetraiodothyronine (T4). The coupling of one monoiodotyrosine and one diiodotyrosine moleculeresults in triiodothyronine (T3).TG is a major autoantigen in autoimmune thyroiditis. Serum antibodies against thyroglobulin may be present at highconcentrations in patients with Graves’ disease and Hashimoto’s thyroiditis.TG autoantibodies are also found in some clinically euthyroid individuals, but usually at lower concentrations than inpatients with clinical disease.In addition to autoimmunity, anti-TG antibodies may develop in patients suffering from thyroid cancer. High levels of anti-TG may interfere with correct determination of serum thyroglobulin. TG is an established tumor

2. INTENDED USE

is intended for the quantitative determination of autoantibodies to TG (thyroglobulin) in human serum or plasma (citrate).
3. PRINCIPLE OF THE ASSAY
The quantitative immunoenzymatic determination of autoantibodies against thyroglobulin (TG) is based on the
ELISA
(Enzyme-linked Immunosorbent Assay) technique. Microtiter strips are precoated with human thyroglobulin (TG) to bind corresponding antibodies of the specimen. After washing the wells to remove all unbound sample material horser

sh peroxidase (HRP) labelled anti-human IgG conjugate is added. This conjugate binds to the captured TG-specific antibodies. The immune complex formed by the bound conjugate is visualized by adding Tetramethylbenzidine (TMB) substrate which gives a blue reaction product. The intensity of this product is proportional to the amount of TG specific IgG antibodies in the specimen. Sulphuric acid is added to stop the reaction. This produces a yellow endpoint colour. microwell plate reader.
